Scatter and Shatter: Centrosome Declustering is an Attractive Chemotherapeutic Strategy
نویسنده
چکیده
The centrosome is a non-membranous organelle often found near the cell center, as per its namesake. It consists of two orthogonallyoriented centrioles enmeshed within a conglomeration of proteins called the pericentriolar material (PCM). Despite its diminutive size, the centrosome assumes a highly influential position in metazoan cells, serving as the major microtubule organizing center (MTOC). As such, the centrosome impacts a vast array of cellular processes, such as mitotic spindle formation and orientation, cell polarization, directional migration, cell signaling through the primary cilium, and locomotion via flagella [1]. Furthermore, the centrosome serves various MTOC-independent functions in cell cycle progression, such as G1/S transition, mitotic entry, anaphase onset, and cytokinesis. Consequently, disturbances in centrosome function typically engender far-reaching consequences.

Identification of griseofulvin as an inhibitor of centrosomal clustering in a phenotype-based screen.
A major drawback of cancer chemotherapy is the lack of tumor-specific targets which would allow for the selective eradication of malignant cells without affecting healthy tissues. In contrast with normal cells, most tumor cells contain multiple centrosomes, associated with the formation of multipolar mitotic spindles and chromosome segregation defects. Many tumor cells regain mitotic stability ...

متن کاملSynthesis, molecular editing, and biological assessment of the potent cytotoxin leiodermatolide.
It was by way of total synthesis that the issues concerning the stereostructure of leiodermatolide (1) have recently been solved; with the target now being unambiguously defined, the mission of synthesis changes as to secure a meaningful supply of this exceedingly scarce natural product derived from a deep-sea sponge. To this end, a scalable route of 19 steps (longest linear sequence) has been ...
متن کاملMulti-Site Declustering Strategies for Very High Database Service Availability
The thesis introduces the concept of multi-site declustering strategies with self repair for databases demanding very high service availability. Existing work on declustering strategies are centered around providing high performance and reliability inside a small geographical area (site). Applications demanding robustness against site failures like fire and power outages, can not use these meth...
متن کاملChained Declustering: A New Availability Strategy for Multiprocessor Database Machines
This paper presents a new strategy for increasing the availability of data in multi-processor, shared-nothing database machines. This technique, termed chained declustering, is demonstrated to provide superior performance in the event of failures while maintaining a very high degree of data availability. Furthermore, unlike most earlier replication strategies, the implementation of chained decl...
